Characterization of a Novel Emaravirus Affecting Ash Species (Fraxinus spp.) in Europe

چکیده

We identified a novel virus in diseased European ash (Fraxinus excelsior) and manna (F. ornus) trees exhibiting chlorotic ringspots, mottle leaf deformation such as curling shoestring symptoms. High-throughput sequencing (HTS, Illumina RNASeq) of total RNA isolated from material combination with RT-PCR-based amplification techniques Sanger determined five complete genome segments, each encoding single open reading frame. Sequence analyses RNA1–RNA5 revealed organization typical for emaraviruses, i.e., (i) conserved complementary terminal 5′ 3′ termini segment (ii) proteins showing significant homologies to the RNA-dependent polymerase (RdRP) encoded by RNA1, glycoprotein precursor (GPP) RNA2, viral nucleocapsid protein (N, RNA3), movement (MP, RNA4), 26 kDA (P26, RNA5) highly similar unknown function other emaraviruses. Furthermore, we spherical particles (double-membrane bodies, DMB) different sizes (70–80 nm diameter) which are emaraviruses exclusively virus-infected tissue deformation. comparison phylogenetic confirmed new member genus Emaravirus. established species-specific RT-PCR detection protocol could associate observed disease symptoms infection emaravirus F. excelsior ornus. Therefore, propose name shoestring-associated (ASaV). Investigation ASaV-infected sample originating locations Switzerland, Germany, Italy Sweden provided wide geographical distribution affected species. To our knowledge, this is first confirmation an affecting tree species leaves Europe.

Transcriptomic Signatures of Ash (Fraxinus spp.) Phloem

BACKGROUND Ash (Fraxinus spp.) is a dominant tree species throughout urban and forested landscapes of North America (NA). The rapid invasion of NA by emerald ash borer (Agrilus planipennis), a wood-boring beetle endemic to Eastern Asia, has resulted in the death of millions of ash trees and threatens billions more. Larvae feed primarily on phloem tissue, which girdles and kills the tree. While ...

Chloroplast DNA variation and postglacial recolonization of common ash (Fraxinus excelsior L.) in Europe.

We used chloroplast polymerase chain reaction-restriction-fragment length polymorphism (PCR-RFLP) and chloroplast microsatellites to assess the structure of genetic variation and postglacial history across the entire natural range of the common ash (Fraxinus excelsior L.), a broad-leaved wind-pollinated and wind-dispersed European forest tree. A low level of polymorphism was observed, with only...

Host selection and feeding preference of Agrilus planipennis (Coleoptera: Buprestidae) on ash (Fraxinus spp.).

We studied the host selection behavior and feeding preference of the emerald ash borer, Agrilus planipennis Fairmaire (Coleoptera: Buprestidae). A. planipennis is an exotic forest insect pest native to Asia that was discovered in North America in 2002 and is causing widespread mortality of ash trees (Fraxinus spp.) in southeast Michigan and surrounding states. We compared host selection and fee...

Genetic Characterization of Common ash (Fraxinus excelsior L.) populations in Bosnia and Herzegovina

Background and Purpose: Common ash (Fraxinus excelsior L.) is a species with a widespread distribution in Europe. It mostly grows on mesophyll sites, mainly in karst valleys and by forest creeks, and in Herzegovina at northern expositions and higher altitudes with somewhat better types of soil. Materials and Methods: We studied 10 natural populations. We collected dormant buds with branches fro...
